Glucose is monosaccharide found in fruits and also derived From the breakdown of carbohydrates in the diet & the conversion of glycogen by liver. Glucose in the body’s main source for cellular energy, brain. Excess glucose is stored as glycogen in the liver and muscle cells. The glucose level in the blood is maintained by insulin (cause lowering The glucose level) and glucagon, adrenalin and cortisol(cause elevation the glucose level).

Renal threshold for glucose is 180mg/dl. Glucoseuria is the presence of glucose in urine, when the blood glucose exceed the renal threshold  for glucose, in pregnancy renal threshold is reduced.

Sample:

Serum or plasma ,free of hemolysis.

Fasting sample 8-10 hr over night ; sample collection :preserve with sodium florid.

Glucose is stable at 2-8 ºc for 3 days.

OGTT : 3 days before the sample collect patient receives diet containing 150g of carbohydrate/day.

Test Principle:

In presence of peroxidase (POD), the formed hydrogen peroxide

formed affects the oxidative coupling of phenol and 4-aminoantipyrine (4-AAP) to form a red-colored quinoneiminedye.
