Uric acid test

-Purine compound produced from break down of nucleic acids present in animal cell nucleolus, uric acid is secreted via kidney & GIT, skin about 70% is secreted via kidneys.

-Uric acid filtered in the form of urate and then partially reabsorbed

-Principle:

Phosphotungestic acid + urate  ==> phosphotungestate (Blue color).

Uric acid + 2H2O +O2  ==>by Uricase ==>  Allantoins +H2O2 +CO2.

Absorbs at 293 nm                                                         not Absorbs at 293 nm

H2O2  react with bizidine dreveitives or other Hydrogen doners to produce achromogen.

Sample :–

Serum, heparinized or EDTA plasma.

Nomal range:–

 Male : (3.5 – 7.2 mg/dl).

Female : (2.6 – 6.0 mg/dl).

Serum Uric acid increased in:–

– Gout.

– Renal failure.

High purine dietary intake.-

Acute leukemia.  –

– Chemotherapy or radiotherapy of large tumors.

-Serum Uric acid decreased in:

– Low purine dietary intake.
